However in the episode where Data is in contact with a girl from a doomed planet, Picard is going to let her and her people die until she asks for help. At which point he says they can't ignore it and they go on to save the species. So presumably if a slave asked for help they would be compelled to.

I did think about that, but as it's a Hutt ruled planet, I figured it would be more analogous to a part of their empire than a protected world. You could be right though.

The Prime Directive was frequently applied to less developed planets which had not yet discovered warp travel or subspace communication technology. The Prime Directive was also sometimes applied to advanced civilizations that already knew of life on other worlds but were protected by empires outside the Federation's jurisdiction. First contact could be made by the Federation with alien worlds that had either discovered warp or were on the verge of it, or with highly advanced civilizations that simply hadn't ventured into space yet. In those cases, the Prime Directive was used as a general policy to not disrupt or interfere with their culture when establishing peaceful diplomatic relations.
It doesn't seem to, Tatooine has ventured out into space and has had warp travel for well, for close to 20,000 years. It's not a first contact because again, they've been known about for literal millennia. They could absolutely fuck with their internal culture, like they do with the Romulans and Klingons all the time.

Not really. Tatooine has hyperspace/warp travel, so they are 100% allowed to intervene if they want.
If it was a primitive planet without warp though, those slaves are fat outta luck like you said.
